The skit was him getting mad at the audience because they weren't asking him questions of substance, they were asking him his fav color and what not. He finally gets fed up and says, "night after night they want a trump joke, or do carpool karaoke with Britney effing spear, I AM DONE!" So I think the joke is that he is tired of doing this crap  when no one appreciates him, or something like that, but that's just my interpretation

It's just a joke. I think the rule is, whenever someone explicitly slags someone else off it's meant to be taken as humour. 

I hate him, but I imagine he was DYING belting out Britney's pop songs with her bopping beside him mouthing the words. As much as it was awkward for her, he must have been so frustrated with that, too.
